Enhanced Enzymatic Production of Cephalexin at High Substrate Concentration with in situ Product Removal by Complexation
نویسندگان
چکیده
Cephalexin (CEX) was synthesized with 7-amino-3-deacetoxycephalosporanic acid (7-ADCA) and D(–)-phenylglycine methyl ester (PGME) using immobilized penicillin G acylase from Escherichia coli. It was found that substrate concentration and in situ product could remarkably influence the ratio of synthesis to hydrolysis (S/H) and the efficiency of CEX synthesis. The optimal ratio of enzyme to substrate was 65 IU/mM 7-ADCA. High substrate concentration improved the 7-ADCA conversion from 61 to 81 % in the process without in situ product removal (ISPR), while in the synthetic process with ISPR, high substrate concentration increased the 7-ADCA conversion from 88 to 98 %. CEX was easily separated from CEX/b-naphthol complex and its purity and overall yield were 99 and 70 %, respectively.

Simultaneous high hydrogen content-synthesis gas production and in-situ CO2 removal via sorption-enhanced reaction process: modeling, sensitivity analysis and multi-objective optimization using NSGA-II algorithm
The main focus of this study is improvement of the steam-methane reforming (SMR) process by in-situ CO2 removal to produce high hydrogen content synthesis gas. Sorption-enhanced (SE) concept is applied to improve process performance. In the proposed structure, the solid phase CO2 adsorbents and pre-reformed gas stream are introduced to a gas-flowing solids-fixed bed reactor (GFSFBR). One dimens...

Recovery of metabolites from fermentation broth by solvent extraction can be used to optimize fermentation processes. End-product reutilization, low product concentration and large volumes of fermentation broth and the requirements for large bioreactors, in addition to the high cost largely contributed to the decline in fermentative 2,3-butanediol production. Extraction can successfully be ...
متن کاملEnhanced Bioethanol Production in Batch Fermentation by Pervaporation Using a PDMS Membrane Bioreactor
The integration of batch fermentation and membrane-based pervaporation process in a membrane bioreactor (MBR) was studied to enhance bioethanol production compared to conventional batch fermentation operated at optimum condition. For this purpose, a laboratory-scale MBR system was designed and fabricated. Dense hydrophobic Polydimethylsiloxane (PDMS) membrane was used for pervaporation. For fer...
متن کاملA Pore Scale Evaluation of Produced Biosurfactants for Ex-situ Enhanced Oil Recovery
Microbial enhanced oil recovery (MEOR) is an economical method used to improve the oil recovery from reservoirs. In the MEOR techniques, by applying different microorganisms, a variety of products such as bioacid, biogas, biosurfactant, and biopolymer are generated, among which biosurfactant, one of the important metabolites, is produced by bacteria. It is worthy to note that bacteria are suita...
